Association between ABCA1 gene 13148 C/T polymorphism and type 2 diabetes mellitus
AIM: To observe whether the ABCA1 gene 13148C/ T single-nucleotide polymorphism is associated with type 2 diabetes mellitus and lipid metabolism. METHODS: The genotype and allele frequencies of ABCA1 13148C/T genetic variation were assayed by nest-polymerase chain reaction-restriction fragment length polymorphism ( nest-PCR-RFLP ). A case-control study was conducted in 148 patients with type 2 diabetes mellitus and 114 healthy controls. RESULTS: The CC genotype frequency in type 2 diabetes mellitus group was lower than that in healthy controls ( P < 0. 05 ) while the TT genotype frequency in type 2 diabetes mellitus group was higher than that in healthy controls ( P < 0. 05),with significant difference in the frequency of the T allele between DM and CON group (P<0.05). The BMI,FPG,SBP,TC,TG,LDL-C,Fins,HOMA-IR and HbA1c levels in type 2 diabetes mellitus group were higher than those in healthy controls (P < 0. 05),while HDL-C in type 2 diabetes mellitus group was lower than that in healthy controls (P < 0. 05 ). The level of TG and HOMA-IR gradually increased while that of HDL-C gradually decreased in genotype of CC,CT and TT. HDL-C level of TT genotype was significantly lower than that of CC genotype (P<0. 05). CONCLUSION:There exists an association between ABCA1 13148C/T polymorphism and type 2 diabetes mellitus,which is associated,to some extent,with lipid metabolism. T allele and TT genotype appear to be risk factors for type 2 diabetes mellitus in Han nationality in Gansu Province of China.
