Effect of specific siRNA targeting cerbB1 on EGFR and survivin expression and cell proliferation in human meningioams
AIM: To study the influence of siRNA targeting cerbBl on cell proliferation and expression of EGFR and survivin in cultured human meningioma cells. METHODS: Small interfering RNA ( siRNA) targeting cerbB1 was constructed and transfected into human meningioina cells in vitro. Flow cytometry and MTT method were used to detect the effect of siRNA on the proliferation and apoptosis of cultured human meningioma cells. The expression of EGFR and survivin was analyzed by Western Blot and RT-PCR after siRNA interference in meningioma cells. RESULTS: The meningioma cell proliferation was inhibited significantly after siRNA interference,the apoptosis and necrosis cells in siRNA group increased significantly,and the expression of EGFR and survivin was significantly down-regulated after interference. CONCLUSION: siRNA targeting cerbBl significantly down-regulates the expression of EGFR and survivin,inhibits the proliferation and induces the apoptosis of meningioma cells. The findings of this study provide some experimental base for the gene targeting treatment of meningiomas.
