Expression of TCTP mRNA and its biological significance in liver regeneration
AIM: To investigate the expression of TCTP mRNA and its biological significance in liver regeneration. METHODS: Liver regeneration model was established by a two-thirds partial hepatectomy(PH) in healthy adult male SD rats. The regenerative liver tissues were harvested at different time-points after PH. The mitotic index(MI) was evaluated under microscope and the distri-bution in G0/G1-, S- and G2/M-phase subpopulation was deter-mined by flow cytometry. The expression of TCTP mRNA was measured by semi-quantitative RT-PCR. RESULTS: After PH, the MI of hepatocytes increased significantly at 3 to 12 h. Cell cycle analysis showed a G0/G1- to S-phase transition at 1 h and an increased proportion of G2/M-phase cells at 3 h, followed by the most significant increase at 6 h. The mRNA expression of TCTP was up-regulated slightly at 1 h, markedly increased at 3 to 12 h and then declined to the original level. CONCLUSION: The dynamic changes of TCTP mRNA expression may be related to the mitosis and cell proliferation in liver regeneration.
