Curative effect of pectinketoprofen prodrug on experimental ulcerative colitis in rats
AIM: To study the curative effect of pectin-keto-profen (PT-KP) prodrug on experimental ulcerative colitis induced by 2, 4, 6-trinitrobenzene sulfonic acid (TNBS) in rats. METHODS: Thirty rats with ulcerative colitis induced with TNBS at the dose of 100 mg/kg were randomized into five groups: ent doses by intragastric administration, while the other groups were given sodium carboxymethyl cellulose(5 mL/L) once a day for 6 d by intragastric administration. The thymus, the spleen and the colon were taken at the end of the experiment. The weights of thymus, spleen and colon were measured and the ratio of organ weight to body weight was calculated. The morphology of rat colons was observed and the colons were photographed. The ulcerative areas were measured and the percents of ulcerative area were calculated by picture processing system on computer. The colons were fixed in 40 g/L formaldehyde and their histopathologic changes were examined. RESULTS: After the oral administration colon were reduced to 30.43% and 5.80% respectively and the ratios of colon weight to body weight were reduced to 5.72 and 3.23 respectively, significantly lower than those in model group. However, no significant effects were found on the weights of thy-CONCLUSION: PT-KP prodrng has a good therapeutic effect onulcerative colitis in rats.
