摘要

目的:建立一种利用高铁氰化钾法检测鸡胚尿囊膜(CAM)血管生成的方法.方法:分离发育期鸡胚尿囊膜并将其匀浆,加入高铁氰化钾溶液,用紫外分光光度计测量A值,通过标准品计算样品中血红蛋白的含量.结果:采用该方法测定了发育7及9 d的鸡胚尿囊膜血红蛋白含量,以及中等强度静磁场对不同发育阶段新生血管的抑制效应,而且与体视显微镜结果一致.结论:高铁氰化钾法是一种实用的可定量检测鸡胚绒毛尿囊膜新生血管的实验方法.

Abstract

AIM: To establish a new method to detect chicken chorioallantoic membrane (CAM) angiogenesis. METHODS: Developing CAM was separated and homogenized. The high potassium ferrocyanide solution was added to CAM homogenates and the absorbanco was detected by ultraviolet spectrophotometer. The hemoglobin content of CAM homogenates was obtained by reference standards. RESULTS: The hemoglobin content of CAM being developed for 7 and 9 d and the inhibitory effects of medium static magnetic field on the hemoglobin content of CAM were detected. The results obtained by detection of the hemoglobin content of CAM were consistent with those by stereo microscope. CONCLUSION: The high potassium ferricyanide method is a practical and quantitative approach to detect the CAM angio-genesis.
