目的:探讨同一生物组织在组织、细胞和细胞膜不同层面的傅立叶变换红外光谱(FTIR)谱图变化规律.方法:将提取肿瘤组织和瘤旁正常组织的细胞膜及提取细胞膜后剩余成份(沉淀物),于100 kg/cm2下8 min制成溴化钾压片行FTIR检测,指认其特征光谱并将细胞膜、沉淀物、细胞株及组织的FTIR谱图进行对比分析.结果:细胞膜与沉淀物的FTIR谱图相比,蛋白和脂类的吸收峰发生显著变化,核酸的吸收峰在细胞膜消失而在沉淀物中增强;细胞膜的FTIR谱图与组织、细胞株相比,细胞膜的蛋白质二级结构发生了明显改变,脂类吸收峰增强;沉淀物的FTIR谱图与组织、细胞株相比,蛋白二级结构发生了明显改变,脂类吸收峰增强,但与组织谱图一致;癌组织中细胞膜、沉淀物及组织水平的相对强度I1460/I1400均小于正常组织.结论:同一组织不同层面的FTIR谱图变化不全一致;肿瘤组织中脂类含量减少,但细胞膜上的脂类含量增加,肿瘤组织及肿瘤组织细胞膜的蛋白质二级结构发生了明显改变;1460 cm-1和1400 cm-1可作为FTIR用于肿瘤诊断的一个可靠指标.
Comparative study on Fourier transform infrared spectroscopy of tissues at different tissue levels
AIM: To investigate the Fourier transform infrared spectroscopy (FTIR) spectrum of the same tissue at the level of the tissue, the cell and the plasma membrane. METHODS: Plasma membrane and remaining ingredients were extracted from cancer tissues and normal tissues, and then compressed into thin KBr disks under a pressure of 100 kg/cm2 for 8 min for FTIR investigation. The spectra obtained from the cell line, tissue, plasma membrane and remaining ingredients were compared. RESULTS: Compared with that in tissue FTIR spectrum, the absorb peak of protein and lipids related bands changed significantly in plasma membrane and remaining ingredients. The absorb peak intensity of nucleic acid related bands disappeared in plasma membrane but increased in the remaining ingredients. The absorb peak intensity of protein and lipids related bands increased in plasma membrane compared with that in tissue and cell line. The absorb peak intensity of protein and lipids related bands increased in the remaining ingredients, similarly as that in tissues. The relative intensity of I1460/I1400 in plasma membrane, remaining ingredients and cancer tissues was weaker than that in the normal tissues. CONCLUSION: The FTIR spectra, obtained from the same tissue but at different tissue levels, are different. The content of lipids decreases in cancer tissues but increases in the plasma membrane. The secondary sturctures of proteins in cancer tissues and plasma membrane change greatly. 1460 cm-1 and 1400 cm-1 can be used as reliable indicators in tumor diagnosis.
