Differentiation of U937 cells induced by Matrine and its mechanism
AIM: To investigate the effects of Matrine on the differentiation in U937 cells and its mechanism. METHODS: The distribution of cell cycle was measured by flow cytometry. Cell differentiation was detected by NBT reduction test, expression of cell surface markers (CD11b and CD14) and morphologic observation. Western blot was used to detect the protein expression of p21Waf1/Cip1. RESULTS: Cell-cycle analyses of the Matrine-treated cells indicated that the cells were arrested in S-phase. The NBT positive cells and the expression of CD11b increased obviously (P < 0.05). The expression of p21Waf1/ Cip1 was significantly up-regulated in Marrine-treated U937 cells (P < 0.05). CONCLUSION: Matrine induces the differentiation of U937 cells, which may be related to the changes of p21Waf1/Cip1.
