苦参碱诱导U937细胞分化及其相关机制

目的:观察苦参碱对U937细胞诱导分化作用,并探讨其可能作用机制. 方法:采用流式细胞仪检测细胞周期分布;硝基四氮唑蓝(NBT)还原实验、CD11b,CD14表面抗原的表达及细胞形态学检测细胞分化;Western Blot检测p21Wafl/Cipl的蛋白表达. 结果:苦参碱作用U937细胞后,细胞周期分析提示发生S期阻滞;NBT阳性细胞率明显增高(P<0.05),CD11b表达增加(P<0.05);细胞内p21Wafl/Cipl表达增高(P<0.05). 结论:苦参碱能诱导U937细胞分化,其机制可能与细胞周期调控蛋白p21Waf1/Cip1表达的改变有关.
Differentiation of U937 cells induced by Matrine and its mechanism
AIM: To investigate the effects of Matrine on the differentiation in U937 cells and its mechanism. METHODS: The distribution of cell cycle was measured by flow cytometry. Cell differentiation was detected by NBT reduction test, expression of cell surface markers (CD11b and CD14) and morphologic observation. Western blot was used to detect the protein expression of p21Waf1/Cip1. RESULTS: Cell-cycle analyses of the Matrine-treated cells indicated that the cells were arrested in S-phase. The NBT positive cells and the expression of CD11b increased obviously (P < 0.05). The expression of p21Waf1/ Cip1 was significantly up-regulated in Marrine-treated U937 cells (P < 0.05). CONCLUSION: Matrine induces the differentiation of U937 cells, which may be related to the changes of p21Waf1/Cip1.
