三甲益肝颗粒含药血清对HSC-T6细胞TGF-β1mRNA和TIMP-1 mRNA 表达的影响

目的:观察三甲益肝颗粒(S-JC)含药血清对肝星状细胞HSC-T6细胞转化生长凶子β1(TGF-B1)mRNA和基质金属蛋白酶组织抑制因子1(TIMP-1)mRNA表达的影响. 方法:不同的剂量的三甲益肝颗粒(1.7,3.4和6.8 g/kg)灌胃大鼠,制备含药血清,作用于HSC-T6细胞48 h,以逆转录定量PCR方法测定其对HSC-T6细胞TGF-β1 mRNA和TIMP-1mRNA表达的影响. 结果:水飞蓟宾组,三甲益肝颗粒低、中、高剂量组的TGF-61 mRNA,TIMP-1 mRNA表达水平分别为1.65±0.05,1.67±0.04,1.23±0.07,0.72±0.02,2.23±0.12,2.17±0.12,1.41±0.03,0.49±0.09,与对照组的TGF-β1 mRNA和TIMP-1 mRNA表达水平(2.11±0.11,2.72±0.11)比较,具有显著性差异(P<0.05). 结论:三甲益肝颗粒降低HSC-T6细胞TGF-β1 mRNA,TIMP-1 mRNA表达水平,从分子水平上证明其抗肝纤维化的作用.
Effect of Sanjiayi Gan granula contained serum on TGF-β1, TIMP-1 gene expression of HSC-T6 cells
AIM: To observe the in vitro effect of Sanjiayi Gan granula (S-JG) contained serum on TGF-β1 mRNA, TIMP-1 mRNA expression of HSC-T6 cells. METHODS: HSC-T6 cells were exposed in different doses of Sanjiayi gan granula contained serum for 48 h. TGF-β1 mRNA, TIMP-1 mRNA levels were measured by quantitative reverse-transcription polymerase chain reaction (RT-PCR). RESULTS: TGF-β1 mRNA, TIMP-1 mRNA levels of HSC-T6 cells at different doses of S-JG contained serum were 1.65 ± 0.05, 1.67 ± 0.04, 1.23 ± 0.07, 0.72 ± 0.02 and 2.23 ± 0.12, 2.17 ± 0.12, 1.41 ± 0.03, 0.49 ± 0.09, respectively, which were lower than those of normal control (2.11 ± 0.11, 2.72 ± 0.11, P < 0.05, respectively). CONCLUSION: Sanjiayi gan granula (S-JG) contained serum can decrease TGF-β1 mRNA, TIMP-1 mRNA expression of HSC-T6 cells.
