目的:检测齐墩果酸(OA)对人肺腺癌A549细胞增殖和细胞周期的作用. 方法:体外培养人肺腺煽细胞系A549,取指数生长期细胞用于实验.将OA按照1.25,2.5,5,10,20,40 mg/L配制成不同终浓度的OA溶液.实验按照如下方法进行:①采用四甲基偶氮唑蓝(MTT)法检测不同浓度OA及空白对照干预24,48,72 h时A549细胞的增殖抑制率;②流式细胞术检测不同浓度OA干预24 h时细胞周期时相的分布. 结果:A549细胞在倒置显微镜下观察可见细胞生长良好、形态正常、增殖活跃,符合实验要求的数量和活性;OA对A549细胞的增殖抑制率随其浓度的增加及作用时间的延长而升高(P<0.05);10,20,40 mg/L OA干预24 h时G0/G1期细胞比例较对照组增加,呈浓度-效应依赖关系(P<0.01),且该比例有随OA浓度增加而升高的趋势. 结论:OA具有时间和浓度依赖性的抗肺腺癌细胞的活性;其对肿瘤细胞的增殖抑制作用可能是通过阻滞肿瘤细胞于G1期而实现的.
Effect of oleanolic acid on proliferation and cell cycle block of human lung adenoma A549
AIM: To study the effects of oleanolic acid (OA) on the cell proliferation and the cycle block in human lung adenoma cell line A549. METHODS: A549 cells were incubated in vitro and the mice were assigned to groups with OA concentration of 1.25, 2.5, 5, 10, 20 and 40 mg/L. The cell proliferation and the proliferation inhibition rate were detected by MTT assay at 24, 48 and 72 h. The distribution of cells in cycle was assessed by flow cytometry (FCM) at 24 h. RESULTS: The proliferation inhibition rates of A549 rose with the increase of OA concentration and time length(P<0.05). At 24h, the cell ratio of the cells in G0/G1 cycles was higher in 10, 20 and 40 mg/L groups compared with that in control. The cell ratio increased in a concentration-effect dependent manner (P < 0.01) and the ratio rose with the increase of OA concentration. CONCLUSION: OA exerts activated effects on anti-lung adenoma cells in a time and dosage dependent manner and inhibits the proliferation of cancer cells possibly as a result of G1 cycle block in the cells.
