Effects of MT1H gene on drug resistance in human lung adenocarcinoma cell line A549
AIM: To study the effect of MT1H gene on drug resistance in human lung adenocarcinoma A549 cells. METHODS: Recombinant eukaryotic expression plasmid pcDNA3.1 ( - )-MT1H was constructed and then transfected into lung adenocarcinoma cell A549 by LipofectamineTM 2000. The positive cell clones were selected by G418. RT-PCR and Western blot were used to measure the MT1H mRNA and protein expression levels respectively. MTT was used to determine the chemosensitivity to cisplatin. FCM was used to measure the cisplatin-induced apoptosis rate. RESULTS: After transfection of MT1H, MT1H mRNA and protein expression levels were enhanced. The sensitivity to cisplatin and cisplatin-induced apoptosis rate were significantly decreased. CONCLUSION: MT1H may promote drug resistance in A549 cells, which may be related to down-regulation of cisplatin-induced apoptosis.
