目的:制备IL-18基因修饰的树突状细胞AFP瘤苗,探索增强肝癌树突状细胞免疫治疗效果的新途径. 方法:构建和制备rAd-IL18-AFP双基因表达的重组腺病毒,转染DC,应用RT-PCR和Western Blot方法检测IL-18和AFP基因在DC中的蛋白表达. 结果:成功制备IL-18基因修饰的树突状细胞AFP瘤苗,rAd-IL18-AFP介导的IL-18和AFP基因能够在DC中进行蛋白表达. 结论:成功制备IL-18基因修饰的树突状细胞AFP瘤苗,为进一步的肝癌免疫基凶治疗研究打下基础.
Construction of ILI8-AFP-DC vaccine mediated by IL-18 and AFP gene coexpressing adenovirus and detection of its expression
AIM: To construct the IL18-AFP-DC vaccine mediated by IL-18 and AFP gene coexpressing adenovirus and to explore a new method of improving the immune response against hepatocellular carcinoma. METHODS: Cytokine interleukin-18 and AFP gene were transfected into dendritic cells (DC) mediated by adenovirus and the expressions of cytokine interleukin-18 and AFP gene in DC were detected by RT-PCR and Western blot. RESULTS: The IL18-DC-AFP vaccine was successful constructed. CONCLUSION: The IL18-AFP-DC vaccine mediated by IL-18 and AFP gene coexpressing adenovirns is successfully constructed, which has laid the basis for the downstream study of improving immune response against hepatocellular carcinoma.
