AIM: To establish a new method to prepare baicalein and to determine the content of baicalein by UV-spectrophotometry. METHODS: Water was used as the extracting solvent of baicalin and enzyme was used to convert baicalin to baicalein. The effects of temperature, pH value, enzyme activity and reaction time were studied respectively and the process conditions were optimized. The detection wavelength was 244 nm and methanol was used as the solvent. The content of baicalein was determined by UV-spectrophotometry. RESULTS: The content of baicalein reached 93.23%. The linear range of determination was 1-24 mg/L. Regression equation; A = 0.0032C (mg/L) + 0.0045, R2 = 0.985. CONCLUSION: The newly established method is convenient, practical and non-polluting. This method is suitable for the preparation and quantitative analysis of baicalein.
