Determination of eplerenone in human plasma by HPLC-MS
AIM: To develop an HPLC-MS method for the determination of Eplerenone concentration in human plasma. METHODS: The samples were extracted with ethyl acetate after adding the internal standard solution, and analytes were detected by mass spectrometer in positive models. RESULTS: The standard curve was linear in the range of 2.080 - 4160 μg/L and the mean calibration curve: f = 0.0005520 + 0.001333 × C (r = 0.9984). The relative standard derivation of intra- and inter-run was all less than 15%. The extraction recoveries of eplerenone were between 73.6% - 75.5%. CONCLUSION: A specific, sensitive and simple method has been established, which is good for determination of eplerenone concentration in human plasma and for pharmacokinetic studies.
