EPHB6受体型酪氨酸蛋白激酶对ERK信号通路的调控效应

目的:探讨促红细胞生成素产生肝细胞B6(EPHB6)对于胞外信号调节激酶(ERK)信号转导通路的作用.方法:以非小细胞肺癌细胞系A549作为研究对象,采用Western Blot法检测ERK的磷酸化水平及通路检测(PathDe-tect)法检测ERK下游分子和Ets结构域蛋白1(Elk-1)转录因子的活性.结果:A549细胞中EPHB6的表达导致ERK的磷酸化.相反,siRNA干涉EPHB6的表达可以逆转ERK的磷酸化.并且,EPHB6导致的ERK磷酸化与Elk-1转录因子脱偶联.结论:表明EPHB6对于ERK信号通路具有调控作用.
Regulation of EPHB6 receptor tyrosine kinase on the ERK signaling pathway
AIM:To investigate the regulatory effects of EPHB6 receptor tyrosine kinase on the signaling pathway of ERK MAPK. METHODS:A549 non-small cell lung cancer cell line was used in the experiments. Western blotting was used to assess the phosphorylation levels of EPHB6, and the status of Elk-1, a transcriptional activator downstream of EPHB6, was determined by PathDetect kit. RESULTS: Expression of EPHB6 induced the expression knocking-down by EPHB6 siRNA of ERK in A549 cells, reversed the phosphorylation of ERK. Moreover, EPHB6-induced phosphorylation of ERK was uncoupled with Elk-1 transcriptional activator. CONCLUSION:EPHB6 plays a role in the regulation of ERK signaling pathway.
