Direct protectory effects of K-opioid receptor agonist U50488H on myocar-dial ischemia- reperfusion in rats
AIM: To study the effects of U50488H (a selective κ-opioid receptor agonist) on the coronary microcirculation and myocardial ultra structure during myocardial ischemia and reperfusion in rats so as to know whether has some direct protective effects. METHODS: Rats were randomly divided into 4 different groups: control group, ischemia-reperfusion group (I/R), U50488H + I/R group, and Nor-BNI + U50488H + I/R group (Nor-BNI, a selective κ-opioid receptor antagonist). The coronary microcirculation and the myocardial ultra structure were determined in rats. RESULTS: The section area of blood capillary in U50488H + I/R group was larger than that in I/R group and Nor-BNI + U50488H + I/R group. The injury of the myocardial ultra structure was significantly more decreased compared with that in I/R group and Nor-BNI + U50488H + I/R group. CONCLUSION: U50488H improves the microcirculation and decreases the injury of myocardial ultra structure through stimulating K-opioid during myocardial ischemia and reperfusion in rats. U50488H has some direct protective effects during myocardial ischemia and reperfusion in rats, but the effect of U50488H can be abolished by Nor-BNI.
