微波诱导鼠成骨肉瘤细胞凋亡的研究

目的:探讨微波诱导鼠成骨肉瘤细胞系UMR 106细胞凋亡及其适宜作用剂量、作用时间.方法:以微波不同剂量和时间作用于鼠成骨肉瘤细胞系,采用形态学观察、MTY法、软琼脂集落形成实验、FCM等方法观察微波诱导成骨肉瘤细胞的凋亡.结果:不同剂量的微波照射大鼠成骨肉瘤细胞后细胞形态发生了改变,随照射剂量的增高,细胞逐渐脱离基质;成骨肉瘤细胞在不同剂量的微波进行相同时间(1 h)照射下存活率均显著下降,并呈剂量依赖性;微波作用于UMR 106细胞的IC50为100 W/m2微波辐射下不同时间后,细胞的存活率显著下降,呈时间依赖性;微波辐射下大鼠成骨肉瘤细胞出现G1期阻滞.结论:微波辐射可诱导大鼠成骨肉瘤细胞凋亡,其最佳作用时间和剂量为1 h和100 W/m2.
Osteosarcoma cells apoptosis induced by microwave
AIM:To elucidate the appropriate dose and time of microwave to induce the apoptosis of UMR 106 osteogenic sarcoma cells. METHODS:Treat the osteogenic sarcoma cells with microwave of different dose and time and use morphology detection, MTT assay, soft agar formation and FCM analysis to examine the apoptosis level. RESULTS: Different dose of microwave caused the morphology change of osteogenic sarcoma cells. With increased dose, more and more cells detached from basal layer. The survival rate of UMR 106 cells decreased a lot following the radiation of microwave with 1 h treatment and demonstrated the dose-dependent trend in MTT assay. The IC50 of microwave radiation on UMR 106 cells was 100 W/m2. FCM analysis showed the G1 arrest of cell cycle in UMR 106 cells under the treatment of microwave radiation. CONCLUSION: Microwave radiation can induce the apoptosis of UMR 106 osteogenic sarcoma cells and the opitimized time and dose were 1 h and 100 W/m2 respectively.
