4个X-STR基因座复合扩增体系的建立及应用

目的:建立DXS9902,DXS7130,DXS9898及DXS9895等4个X染色体特异性短串联重复(X-chromosome specific short tandem repeat,X-STR)基因座的复合扩增体系,调查其多态性并探讨在法医学实践中的应用.方法:复合扩增DXS9902,DXS7130,DXS9898及DXS9895 4个基因座,ABI PRISM 3100进行毛细管电泳检测,Genescan及Genetyper 软件进行检测结果分析及等位基因分型.结果:在中国北方汉族200名无关男性个体及200名无关女性个体中,4个基因座分别发现了6,12,8,6个等位基因.结论:这4个基因座的复合扩增系统有较高的多态性信息,在个体识别和亲权鉴定中有较好的实用价值.
Development and forensic application of 4 X chromosome STR markers typing system
AIM: To devise a multiplex PCR system of DXS9902, DXS7130, DXS9898 and DXS9895 and study the genetic polymorphism and application in forensic science. METHODS: DXS9902, DXS7130, DXS9898 and DXS9895 were amplified simultaneously using a multiplex system and were typed with ABI prism 3100 Genetic Analyzer by Genescan and Genetyper Analysis Softwares. RESULTS: A total of 200 male and 200 female individuals from North China Han population were tested; 4 locus showed respectively 6, 12, 8, 6 alleles. CONCLUSION: The 4 X-STR multiplex system presented high polymorphism and values for forensic identification and paternity testing.
