Effect of sodium ferulate on Caspase-3 and Bcl-2 expression levels of smooth muscle cells in pulmonary arterv of hypoxic pulmonary hypertensive rats
AIM: To investigate the impact of sodium femlate on Caspase-3 and Bcl-2 of smooth muscle cells in pulmonary arterioles of hypoxic pulmonary hypertensive rats. METHODS: Twenty-one male Wistar rats were randomly divided into control group(n = 7), hypoxic group(n =7) and sodium ferulate treated group(n = 7). Mean pulmonary arterial pressure (mPAP) was measured by a right cardiac catheterization. Dry weight of right ventricle(RV), left ventricle with septum(LV + S) were determinded and RV/( LV + S) ratio was calculated. The index of wall thickness of pulmonary arterioles which are the percentage of vascular wall thickness/vascular external diameter (WT%) and the percentage of vascular wall area/total vascular area (WA%) was measured by a computerized image analyzer. The distribution of Caspase-3 and Bcl-2 in smooth muscle cells of pulmonary aterioles were observed by streptavidin peroxidase method (SP), the morphologic changes of small pulmonary arteries and the integrated optical density (IOD) of Caspase-3 and bcl-2 were determined by image analysis technique. The comparisons of the IOD in VSMC in pulmonary vessels among three groups were made to assess the remodeling of small pulmonary arteries. RESULTS: ①In comparison with the control group, the mPAP of rats had been obviously elevated after hypoxia (P < 0.05), as well as WT%, WA% of pulmonary arterioles and RV/(LV + S) (P < 0.05). The mPAP, WT%, WA% of pulmonary arterioles and RV/(LV + S) were much lower in rats of hypoxia with sodium ferulate group than those of hypoxia group (P < 0.05). ②The expression level of Caspase-3 in hypoxia group was lower than that in hypoxia + sodium ferulate group (P < 0.05), the expression level of bcl-2 in hypoxia group was higher than that in hypoxia + sodium ferulate group (P < 0.05). There were no significant difference between the control group and the hypoxia group (P > 0.05). CONCLUSION: Sodium ferulate can increase Caspase-3 and decrease bcl-2 of PASMC.
