目的:建立N-甲基-N-亚硝基脲(N-methyl-Nnitrosourea,MUN)膀胱灌注诱发的SD大鼠膀胱肿瘤动物模型,检测其肿瘤HER-2表达情况.方法:50只SD大鼠随机分两组,对照组膀胱灌注生理盐水,实验组膀胱灌注MNU,3 wk/次,共4次,MNU总量为10 mg;于第1次灌注后第11周切开膀胱取标本,以光镜、免疫组化及RT-PCR为观察手段,对其诱发的膀胱肿瘤进行检测.结果:第11周对照组无肿瘤,实验组诱发膀胱肿瘤率95%,其组织学形态及病理特征与人膀胱肿瘤相似,免疫组化显示部分肿瘤HER-2蛋白阳性表达,RT-PCR显示其HER-2 mRNA阳性.结论:MNU膀胱灌注可成功建立HER-2阳性表达的膀胱肿瘤动物模型.
Preparation of SD rat animal model of bladder tumor and its testing of HER-2
AIM: To establish an animal model of bladder cancer in SD rats by perfusing N-methyl-N-nitrosourea into bladder, and test its expression of HER-2. METHODS: Fifty SD rats were randomly divided into experimental group and control group, the experimental group perfused with MNU(2.5 mg everytime, one time every 3 weeks, 4 times in all) into bladder, the control group with physiological saline solution. The bladders were slited and the biopsies were gotten respectively at the 11th week. Immunohistochemical SP method was applied to detect HER-2 protein of tumor tissues and RT-PCR method was applied to detect HER-2 mRNA. RESULTS: There wasn't any tumor in control group, but the experimental group showed bladder cancers in 95% SD rats. Its morphology and histological features were similar to human bladder cancer. Immunohistochemistry and RT-PCR showed parts of HER-2 protein and HER-2 mRNA were positive. CONCLUSION: We successfully established a HER-2-positive animal model of bladder tumor perfusing bladder with MNU.
