目的:通过对纳米羟基磷灰石根管充填材料与传统根管充填材料(充填用复方麝香草酚散甲醛甲酚复合制剂)进行细胞毒性、根管密合性的对比研究,评价纳米羟基磷灰石材料的细胞毒性、根管壁密合度.方法:①细胞毒性实验:体外培养成骨细胞,添加纳米羟基磷灰石材料浸提液,并以培养液为阴性对照组,充填用复方麝香草酚散甲醛甲酚复合糊剂为阳性对照组.根据ISO标准,采用MTT法进行体外细胞毒实验,测试不同浸提时间的材料浸提液与成骨细胞接触1,3,5,7 d的A值,以评价其细胞毒性.②根管密合性实验:将10颗新鲜拔除的单根牙随机分成两组,每组牙齿按常规方法进行根管预备.实验组为400 g/L纳米羟基磷灰石糊剂充填,对照组为充填用复方麝香草酚散甲醛甲酚复合糊剂充填.拍X线片以保证各组中实验离体牙牙根均完善充填,充分硬固后,进行微渗漏试验.改良葡萄糖检测微渗漏分析装置进行根管密合度实验,30 d后结果由葡萄糖定量分析仪测定.结果:①细胞毒性实验:实验组与阴性对照组无统计学差异,实验组与阳性对照组有统计学差异(P<0.05).②根管密合性试验:对照组葡萄糖浓度明显高于实验组,有统计学差异(P<0.05).结论:纳米羟基磷灰石根管充填材料体外细胞毒性实验阴性,有良好的生物相容性.纳米羟基磷灰石根管充填材料根管壁密合度明显优于充填用复方麝香草酚散甲醛甲酚复合糊剂.
Nanometer hydroxyl apatite root canal backfill material basic research
AIM: Through (backfill disperses formaldehyde cresol compound preparation to the nanometer hydroxyl apatite root sufficient material and the traditional root canal backfill material with compound prescription muskdeer vanilla phenol) to conduct the cell toxicity, the root canal are closed the contrast research. Appraisal nanometer hydroxyl apatite material cell toxicity, root canal wall being closed. METHODS: ①Cell toxicity experiment: In vitro raise osteoblast, the increase nanometer hydroxyl apatite material leaching solution, and take the nutrient fluid as the negative control group, fills disperses the formaldehyde cresol compound cataplasma with the compound prescription muskdeer vanilla phenol for the masculine control group. Uses the MTT law according to the ISO standard to carry on in vitro cytotoxin experiment, the test soaks differently proposes the time the material leaching solution and the osteoblast contacts 1, 3, 5, 7 d the A values, appraises his/her the cell toxicity. ②The root canal is closed the experiment: Divide into 2 groups stochastically 10 new wiping out single rooted teeth, each group of toeet carry on the root canal preparation according to the conventional method. The experimental group is 400 g/L nanometer hydroxyl apatite cataplasma backfill, the control group disperses the formaldehyde cresol compound cataplasma backfill for the backfill with the compound prescription muskdeer vanilla phenol. Makes X piece to guarantee in each group tests the exsomatize tooth root of a tooth to consummate fills, after hardens fully, carries on the micro leakage experiment. The improvement glucose examines the micro leakage analysis installment to carry on the root canal being closed experiment , after 30 d the result determines by the glucose quota analyzer. RESULTS: ① Cell toxicity experiment: The experimental group and the negative control group non-statistics difference, the experimental group and the masculine control group have statistics difference(P < 0.05). ②The root canal is closed the experiment : The control group glucose density is higher than the experimental group obviously, has statistics difference (P < 0.05). CONCLUSION: The nanometer hydroxyl apatite root sufficient material in vitro cell toxicity experiment negative, has the good biological compatibility. Nanometer hydroxyl apatite root sufficient material it surpasses the backfill with the root canal wall being closed aspect to disperse the formaldehyde cresol compound cataplasma obviously with the compound prescription muskdeer vanilla phenol.
