白藜芦醇对体外培养的大鼠皮层神经元谷氨酸神经毒性的干预作用

目的:研究白藜芦醇(Res)对体外培养皮层神经元的影响;探索谷氨酸对皮层神经元的损伤作用及Res对抗谷氨酸损伤的机制.方法:体外培养新生大鼠大脑皮层神经元,实验组加入20 μmol/L的Res,在不同时间点观察Res对培养的皮层神经元的作用,应用MTT法检测神经元存活率作为反应指标;通过在培养基中加入谷氨酸造成皮层神经元兴奋毒性损伤模型,加入最适剂量的Res,观察Res对上述损伤的作用;通过培养细胞形态学观察、MTT法测定存活细胞数及Bax免疫细胞化学染色检测指标来反映谷氨酸对神经元的损伤作用,同时观察Res对抗损伤的机制.结果:20 μmol/L Res对体外培养的皮层神经元的存活率无明显影响;谷氨酸对皮层神经元产生强烈的兴奋毒性,导致大量神经元死亡,MTT结果表明神经元存活率明显下降;免疫组化染色表明谷氨酸可诱导神经元大量表达Bax,促进神经元凋亡;Res可对抗谷氨酸损伤而保护神经元,它可减轻谷氨酸的神经元兴奋毒性,提高神经元的存活率,同时抑制谷氨酸引发的Bax表达上调.结论:适宜剂量的Res对体外培养的皮层神经元无毒副作用,对神经元的存活率无影响,Res可以通过调节Bax的表达对抗谷氨酸兴奋毒性而保护神经元.
Effects of resveratrol on the cortical neuron suffered from glutamate neurotoxicity
AIM:To investigate the effects of resveratrol on the primary cultured cortical neuron,the mechanism of glutamate neurotoxicity and the influence of resveratrol On the glutamate neurotoxicity.METHODS:We observed the effect of resveratrol at different time points by adding 20 μmol/L resveratrol into the culture medium of primary cultured cortical neurons in vitro.Glutamate was added into the culture medium to induce the neurotoxicity.Resveratrol was added to the culture medium simultaneously with glutamate in order to observe its effects on neurotoxicity.Morphological observation,MTT test,as well as Bax immunocytochemistry staining were performed to evaluate both the glutamate neurotoxicity and the protective effects of resveratrol.RESULTS:20 μmol/L resveratrol had no obvious effects on the livability of cultured cortical neurons in vitro.1 mmol/L glutamate could induce so intense neurotoxicity that many neurons died and the cell livability was greatly decreased.Immunocytochemistry staining indicated that the apoptosis promoting protein Bax was up-regulated by glutamate.20 μmol/L resveratrol could partly inhibit the neurotoxicity of glutamate by preventing neuron from dying;increasing the cell livability and down-regulating the expression of apoptosis promoting protein Bax.CONCLUSION:Ahhough resveratrol(20 μmol/L)had neither positive nor side effects on the primary cultures of cortical neuron,it could inhibit the glutamate neurotoxicity and protect neuron by down-regulating the expression of Bax.
