青藤碱对高糖处理下的血管内皮细胞功能的影响

目的:研究青藤碱(SN)对高糖处理的脐静脉血管内皮细胞(HUVECs)NF-KB和NO表达及细胞活力和凋亡的影响.方法:从新鲜脐带中分离培养HUVECs,观察不同浓度SN干预高糖处理的HUVEC的NO,NF-κB的表达以及细胞活力和凋亡的变化.结果:与基础状态相比较高糖可导致HUVECs释放NO改变、NF-κB的表达上调,细胞活力下降,细胞凋亡率上升,而SN在适当浓度可干预上述变化.结论:SN可抑制高糖诱导的脐静脉内皮细胞的损伤和凋亡.
Effects of Sinomenine on function of endothelial cells in medium with high concentration of glucose
AIM:To investigate the effect of Sinomenine on the expression of nitric oxid(NO),nuclear factor kappaB(NF-κB)and proliferation and apoptosis by high glucose in cultured humanumbilical vein endothelial cells(HUVEC).METHODS:HUVECs were isolated from freshly collected umbilical cords.Effect of SN at different concentration on the expression of NO,NF-κB and proliferation and apoptosis of HUVEC cultured with high glucose were observed.RESULTS:Compared with the control group,high glucose could inhibit the proliferation and induce apoptosis,and increase nuclear factor kappaB expression and decrease NO production of HUVECs.Sinomenine could increase the proliferation and decrease the apoptosis,and down nuclear factor kappaB expression and enhance NO production in HUVECs.CONCLUSION:Sinomenine could inhibit the endothelial dysfunction and apoptosis induced by high glucose.
