Association of polymorphism of IL-10 and Helicobacter pylori infection to gastric cancer
AIM:To analyze the relationship both-592 site A/C polymorphism in interleukin-10(IL-10)gene and different virulence factors of Helicobacter pylori in a population from Gansu Province and susceptibility to gastric cancer.METHODS:Polymerase chain restriction fragment length polymorphism (PCR-RFLP)were performed to analyze the genotype of the A/Cpolymorphism in its-592 site of promoter region,and Immunoblotting was performed to test H pylori.RESULTS:①The distribution frequency of AA,AC,CC three genotypes were 29.9%,47.9%,22.2%respectively in normal control.②The frequency of CC genotype was conspicuous higher in the gastric cancer (GC)group compared to the control group(P<0.05),moreover,individuals with the IL-10AC+CC genotype increased 2.19 fold risk for gastric cancer.③Take IL-10 the AA genotype and the H.wlori-immunoblotting negative individual as the comparison,there was an obviously increased risk of developing gastric cancer for those who carried both CC genotype and H.pylori seropositive,CagA positive,increased 3.75,4.51 fold risk respectively.CONCLUSION:Results indicated that there was a relationship between IL-10-592 A/C polymorphism and susceptibility to gastric cancer.And there were statistically interaction between the-592 site A/C polymorphism in IL-10 and H.pylori infection in the occurrence and development process of gastric cancer.
