Study of total flavonoid from fructus sophorae on lipid-lowering in hyperlipidemic rats and its antioxidant capacity
AIM: To explore the antioxidation and the serum lipid lowing effects of total flavonoid from fructus sophorae( TFFS) on experimental hyperlipidemic rat. METHODS: To establish hyperlipoidemic rats model with high lipid diet for 4 weeks and then give experimental treatment with TFFS with dose of 40,80, 120 mg/kg of body weight for 6 weeks. Gypenosides was also used as a positive control, the serum were collected to measure the total cholesterol (TC ) , triglyceride(TG) , high density lipoprotein cho lesterol ( HDL-C), low density lipoprotein cholesterol ( LDL-C ), whole blood viscosity was measured, and the liver tissue was also collected for determining malonaldehyde ( MDA) content and the activities of superoxide dismutase ( SOD) and total antioxidation capacity (T-AOC). The expression level of low density lipoprotein receptor( LDL-R) gene mRNA in liver were determined by re versed transcription polymerase chain reaction( RT-PCR) method. RESULTS: Compared with model group, TFFS could decrease TC, TG and LDL-C level( P <0.05 or P < 0.01) , increase HDL C level ( P < 0. 05 ) in blood, They had the effects of decreasing the whole blood viscosity and the secretion of MDA, increase the activities of SOD and T-AOC, and up-regulation of the expression level of LDL-R gene. CONCLUSION: TFFS has the activity of modulating lipid metabolite of high-lipid rat. Its mechanism may be related with recede blood viscosity, anti-lipoperoxidation and up-regulation of the expression of LDL-R gene in mRNA level.
