RNAi-mediated knockdown of HYAL2 gene inhibits proliferation and invasion in human ovary carcinoma SKOV3 cells
AIM: To study the effect of RNA interference against HYAL gene on the proliferation and invasion in ovarian epthelial carcinoma SKOV3 cell line. METHODS: The transcription of HYAL gene was detected by RT-PCR; the proliferation was analysed by MTT-assay and flow cytometric; penetrating matrigel capacity of the SKOV3 cells was detected by the pattern of vitro invasion. RESULTS: After RNA interference against HYAL gene for 48 h, HYAI2 mRNA was downregulated; the proliferation of the SKOV3 cell was inhibited obviously; flow cytometry showed that cells in S phase were reduced ( P < 0. 05), with more cells arresting in G0/G1 phase of cell cycle ( P < 0. Penetrating matrigel capacity inhibited significantly (P < 0. 05). CONCLUSION: The capcity of proliferation and invasion can be inhibited by RNA interference against HYAL gene. It indicates that HYAI2 gene is likely to become new potential target of gene therapy approach to ovarian epithelial carcinoma.
