C-fos expression in cornu dorsale medullae spinalis in PDLLA/FK506 compound condit peripheral nerve regeneration model in rats
AIM:By observing pain threshold and changes of c-fos expression in the dorsal horn in PDLLA/FKS06 compound condit peripheral nerve regeneration model to investigate regeration nerve's neuropathic pain.METHODS:Forty male Wistar rats weighing 180-200 g were randomly divided into 2 groups:PDLLA/FK506 compound condit regeneration model group(group A)and sham-operation group(group B).The fight sciatic nerve of group A underwent transection and then repaired with PDLLA/FK506 compound condit while 10 mm nerve defect was kept.The right sciatic nerve was just exposed in B group without injury. Behavioral response,pain threshold and thermal withdrawal latency were assessed on 27,30,33,36,39,42 and 45 d after operation.C-fos expression Was exmianed immunohistochemistry on 27,33,39 and 45 d after operation.RESULTS:The cumulative pain scores of group A were higher than those of group B at every time point(P<0.05),while no significant difference existed between each time point in group A.The count of neurons with positive Fos protein in cornu dorsale medullae spinalis L4-5 of group A was higher than that ofgroup B(P<0.05),and significant difference Was found between the c-fos expression level on 27 d compared with that of45 d after operation in group A(P<0.05).As for pain threshold and TWL.more strick contrast between the value of27 and45 d after opration exist in group A compared with that in group B(P<0.05),and significant difference Was found between the level on 27 d compared with that of45 d after operation in group A(P<0.05).CONCLUSION:The regeneration of injured peripheral nerve may induce the c-fos expression and also hyperpathia and changes in pain-related behavior in PDLLA/FKS06 compound condit peripheral nerve regeneration model.
