Effect of oxygenator on expression of lung tissue iNOS and eNOS during acute lung injury in rabbits
AIM:To investigate the effect of of oxygenator on expression of lung tissue iNOS and eNOS during acute lung injury in rabbits and explore its protective mechanism.METHODS:Forty-five rabbits were divided into 3 groups:control group(group A),ALI group(group B)and oxygenator treatment group(group C)with 15 in each group.ALI model was established by special equipment in group B and C,and oxgenator was applied in group C.Blood samples were taken for blood-gas analysis at 0,1,2,3,4 h after trauma.Four hours later,the animals were killed,and the expression of iNOS and eNOS in lung tissue were tested by immunohistochemistry,the weight to dry lung weight ratio,the activities of serum nitric oxide(NO)and lung malondialdehyde(MDA、and myeloperoxidase(MPO)content were determined.RESULTS:Compared with group A,PaO_2,SpO_2 and PaCO_2 significantly decreased(P<0.05).expression of lung tissue iNOS significantly increased while expression of eNOS were significantly decreased (P<0.05),and lung W/D.the activities of serum NO and lung MDA and MPO were significantly increased(P<0.05)in group B.In group C,compared with group B,PaO_2、SpO_2 and PaCO_2 significantly increased(P<0.05),expression of lung tissue iNOS significantly decreased while expression of eNOS were significantiv increased(P<0.05),and lung W/D.the activities of serum NO and lung MDA and MPO were significantly decreased(P<0.05).CONCLUSION:Protective effect of oxygenator on acute lung injury is related to the inhibition of iNOS expression and the inducement of eNOS expression.
