Effect of heat shock protein 27 on apoptosis of spinal cord anterior horn after brachial plexus avulsion in rats
AIM:To observe the effect of heat shock protein 27 on apoptosis of spinal cord anterior horn after brachial plexus roots avulsion.METHODS:Sixty adult Wistar male rats were divided into control and experiment group at random. The experiment group subjected to heat shock under 45℃ for 15 min,and mainrained under 42℃ for 20 min subsequently. After recovering 24 h under the room temperature, the nerves of brachial plexus were avulsion with microhemostatic forcep. In a span from 12 h to 7 d,these animals were killed at different time. But the control group only received the surgery of the nerveroots of brachial plexus avulsion.Apoptosis was detected by TDT-mediated dUTP nick end labeling(TUNEL),the immunohistichemical method wag used to detect leveling the expression of Hsp27.RESULTS:①The hsp27 begin to express at 12 h and significantly increased the level with a peak at 1 d in two group,But the control group contained a lower level in the expression of hsp27 than the experiment group.②TUNEL results showed the peak of apoptosis at 1 d in experiment groups, and in control groups, the peak of apoptosis at 3 d.But the number of apoptosis in experiment group were less than the control group.CONCLUSION:Hsp27 inhibited apoptosis of spinal cord anterior horn motoneuron after braehial plexus roots avulsion.
