恒磁场对阿霉素杀伤人乳腺癌细胞的协同作用

目的:观察恒磁场联合阿霉素对人乳腺癌细胞MCF-7增殖和凋亡的影响及其机制.方法:MCF-7细胞经不同浓度的阿霉素和/或恒磁场(10 mT)处理24 h后.以MTT法检测不同条件下细胞生长受抑制情况;流式细胞术检测各组细胞周期分布;荧光显微镜检测细胞内活性氧变化情况;蛋白电泳检测凋亡相火蛋白Caspase 3表达情况.结果:恒磁场可明显增强阿霉素对MCF-7细胞生长的抑制效应(P<0.01),更强地抑制细胞DNA合成及有丝分裂(P<0.05),促使细胞内活性氧合成,并上调凋亡相关蛋白Caspase-3的表达.结论:恒磁场(10 mT)对阿霉素杀伤人乳腺癌细胞具有较强的协同作用.
Synergistic inhibitory effects of static magnetic fields and adriamycin on breast cancer cell line MCF-7 in vitro
AEM:To explore the synergistic effects of static magnetic fields(SMF)and adriamycin(ADM)on cell growth in human breast cancer cell MCF-7 in vitro.METHODS:The MCF-7 cells were exposed to SMF in combination with various concentrations of ADM for 24 h.To analyze SMF effects in cell growth,MTT assay,flow cytometry,fluorescence microscopy and Western Blot analysis were used.RESULTS:SMF exposure significantly enhanced the cell growth inhibition and apoptosis by ADM,in which cell cycle arrest,cleaved Caspase-3,and reactive oxygen species(ROS)increased.CONCLUSION:SMF(10 mT)could enhance the antitumor effects of ADM,and would be a possibility of comprehensive therapy for breast cancer.
