目的:研究体外培养条件下的弓形虫培养上清对卵巢癌细胞SKOV-3细胞增值的影响及诱导其凋亡的情况.方法:取对数生长期的SKOV-3细胞(数量为5×10~7/L)分别接种于不同细胞培养板,对照组加入RPMI-1640孵育,试验组加入相同体积不同数量(3×10~(10)/L,6×10~(10)/L,12×10~(10)/L)弓形虫速殖子培养上清,四甲基氮噻唑蓝(MTT)法检测吸光度(A_(490nm)值)并计算SKOV-3细胞增值抑制率;Annexin-V-FITC/PI染色细胞后上流式细胞仪检测各个时间点细胞凋亡率;并以荧光显微镜观察SKOV-3细胞形态改变情况;以琼脂糖凝胶电泳观察SKOV-3细胞凋亡DNA条带.结果:MTT法检测结果显示SKOV-3细胞增殖抑制率随着弓形虫速殖子培养上清数量和作用时间增加均明显增大,12×10~(10)/L数量组72 h抑制率最高可达(33.52±2.63)%,弓形虫速殖子培养试验组抑制率与对照组之间比较差异均有统计学意义(P<0.05).弓形虫培养上清流式细胞仪检测显示SKOV-3细胞凋亡率随着时间增加有升高趋势,48 h达到凋亡最高值(18.23%),48 h后凋亡率开始下降,死亡率增加.荧光显微镜观察到试验组培养不同时项的SKOV-3细胞出现不同时期凋亡特征性改变.琼脂糖凝胶电泳见DNA梯形条带.结论:弓形虫速殖子培养上清对体外培养SKOV-3细胞增殖有明显的抑制作用,并可诱导SKOV-3细胞凋亡.
Effect of culture supernatant of Toxoplasma gondii on human ovarian cancer cell line SKOV-3
AIM:To investigate the inhibitive effects of cuhure supernatants of Toxoplasma gondii on the proliferation of human ovarian cancer cell line SKOV-3 and/or its ieduced apoptosis in vitro.METHODS:SKOV-3 cells(concentration of 5×10~7/L)were harvested in the different cell culture plates.The cells were treated for different hours with different concentrations of Toxoplasma gondii culture supernatant(the concentration of tachyzoites was 3×10~(10)/L,6 ×10~(10)/L,12×10~(10)/L respectively).Growth inhibition rates were measured with the MTY method.Apoptosis of the SKOV-3 cells was detected with the following.FCM was used to test the rates of apoptosis of the cancer cells stained with the fluorescent dye of Annexin-v-FITC/PI,fluorescence microscopy was used to observe the changes of eeUular morphology,the agarose electrophoresis was used to detect the DNA changes of SKOV3 cells.RESULTS:The culture supernatant of Toxoplasma gondii inhibited the proliferation of SKOV-3 cells and The growth inhibition rates were increasing with the time and the concentrations,The higheat rate can reach(33.52±2.63)%at the 48 th hour when the concentration was 12×10~(10)/L.The classic cellular morphology of apoptosis in the treated SKOV-3 ceHs were observed under the fluorescence microscope.DNA fragments in the treated cells were revealed by the agarose electrophoresis.In the flow cytometric analysis,a maximum apoptosis rate of 18.23% was detected at 48 h of treatment with the supernatants,but the apoptosis rates decreased and dead rate increased at the treatment time beyond 48 h.CONCLUSION:The culture supematant of Toxoplasma gondii reveals an inhibition to the proliferation of SKOV-3 cells in vitro,and induces their apoptosis in vitro.
