目的:研究CD44~+/CD24~-表型在乳腺癌中的表达并探讨其与乳腺癌各分子亚型的相关性.方法:根据免疫学标志物(ER,PR,HER2,CK5/6,P53)把217例乳腺癌划分为5类分子亚型:basal-like型、Luminal A型、LuminalB型、Her2过表达型和Normal breast-like型.应用双重免疫组织化学检测CD44/CD24双染的情况,分析CD44~+/CD24-表型与乳腺癌5个分子亚型的相关性.结果:217例乳腺癌病例中,luminalA 型130例、luminalB型15例、HER2过表达型21例、basal-like 型29例、Normal breast-like型22例.38%的乳腺癌组织表达CD44~+/CD24~-表型,CD44~+/CD24~-表型在basal-like型、Lu-minal A型、Luminal B型、Her2过表达型、Normal breast-like型乳腺癌中表达率分别为69.0%,36.2%,26.7%,19.0%,31.8%.CD44~+/CD24~-表型在乳腺癌各分子亚型巾的表达具有显著差异性(P=0.003),高表达在basal-like乳腺癌中(69%).结论:CD44~+/CD24~-乳腺癌干细胞与basal-like乳腺癌有相关性;其他乳腺癌干细胞标志物仍需得到进一步鉴定.
Expression and disposition of CD44~+/CD24~-phenotype in molecular subtypes of breast cancer
AIM:To study the expression of CD44~+/CD24~-phenotype and its association with the molecular subtypes of breast cancer in breast cancer. METHODS:Using immunostaining ER, PR, HER2, CK5/6 and P53 to classify the material into 5 molecular subgroups:basal-like subgroup, Luminal A subgroup, Luminal B subgroup, Her2 over-expressing subgroup and Normal breast-like subgroup. Double-staining immunohistochemistry (IHC)was applied for the detection of CD44~ +/CD24 ~-cells. The correlations of the expression of the CD44 ~+/CD24~-to molecular subtypes of breast cancer were analyzed. RESULTS:In 217 cases of breast cancer, basal-like subgroup:29 cases, Luminal A subgroup:130 cases, Luminal B subgroup:15 eases, Her2 overexpressing subgroup:21 cases, Normal breast-like subgroup:22 eases. The expression rates of CD44 ~+/CD24~-in total breast cancer, basal-like subgroup, Luminal A subgroup, Luminal B subgroup, Her2 over-expressing subgroup, Normal breast-like subgroup were 38%, 69%, 36.2%, 26.7%, 19%, 31.8%. The expression rates of CD44 ~+/CD24~-phenotype was high in basal-like breast cancer, which was significantly different from the other subgroups (P = 0. 003). CONCLUSION:The study suggests that the CD44 ~+/CD24~-breast cancer stem cells in tumor tissue is associated with basal-like cancer, and other breast cancer stem cell markers remain to be identified.
