Analysis of Rumen Bacterial Diversity in Jinjiang Cattle Using MiSeq Sequencing Technology
The present study was designed to reveal the rumen bacterial diversity in Jinjiang cattle using MiSeq high-flux sequencing technology.Ruminal liquid samples were obtained from three Jinjiang cattles ( bull ) weighted (400±20) kg with permanent rumen cannulas.Total DNA was extracted and the 16S rDNA V4 to V5 of bacteria was sequenced by MiSeq sequencing technology.The abundance, distribution and α-diversity index of species were analyzed.The results showed that a total of 56 763 valid 16S rDNA sequences and 946 operational taxonomic unit ( OTU) across 3 samples were obtained.The richness indexes, Chao and Ace inde-xes, of ruminal bacterial were 836 and 841, respectively; and alpha diversity indexes, Shannon and Simpson indexes were 4.96 and 0.021 5, respectively.Fifteen phyla, 23 classes, 26 families and 44 genera were identi-fied via taxonomic summary.The dominant phyla among them were Bacteroidetes ( 64.37%) , Firmicutes (21.20%) and Proteobacteria (7.59%).The most abundant classes were Bacteroidia (52.88%), Clostridia (17.03%), and Prevotellaceae (36.26%).Ruminococcaceae (9.33%) and Lachnospiraceae (4.89%) were the predominant family.At genus level, the high abundance of genera were Prevotella (28.97%), Paraprevo-tella (3.08%), Rikenella (2.28%), Clostridium Ⅳ (1.77%), Succiniclastium (1.62%) and Ruminococcus ( 1.53%) .In conclusion, the diversity of rumen bacterial community in Jinjiang cattle is low.Bacteroidetes is the predominant phylum and Firmicutes is the second most abundant phylum.Prevolla is the most abundant ge-nus in the rumen bacterial community.
