为了研究葡萄糖对杂交石斑鱼受精卵孵化、卵黄囊仔鱼乙酰辅酶A羧化酶(ACC)和脂肪酶合成酶(FAS)活性的影响,在杂交石斑鱼(褐点石斑鱼♀×清水石斑鱼♂)受精卵孵化水体添加不同浓度[0(对照组)、2、4、6、8和10 mg/L]的葡萄糖,测定孵化率及畸形率,并测定1、2、3日龄卵黄囊仔鱼全长、ACC和FAS活性的变化.结果表明:葡萄糖浓度对受精卵孵化率及初孵仔鱼畸形率均有显著影响(P<0.05);葡萄糖浓度与受精卵孵化率、初孵仔鱼畸形率的相关性均可用一元二次方程描述,通过回归方程计算,获得最大受精卵孵化率的葡萄糖浓度为3.8 mg/L.葡萄糖浓度低于6 mg/L时,随着葡萄糖浓度的增加,受精卵孵化率随之升高,初孵仔鱼畸形率随之降低,卵黄囊仔鱼全长随之增加.对于1~3日龄仔鱼,葡萄糖添加组ACC及FAS活性均显著高于对照组(P<0.05);随着葡萄糖浓度的增加,在1日龄仔鱼阶段,ACC和FAS活性均呈上升的变化;在2~3日龄仔鱼阶段,ACC活性呈“上升—下降”的变化,FAS活性呈“上升—平缓”的变化.综上分析,在杂交石斑鱼人工育苗孵化水体中添加6 mg/L葡萄糖可显著促进其受精卵孵化及卵黄囊仔鱼发育,显著提高卵黄囊仔鱼ACC及FAS活性.
Effects of Glucose on Fertilized Eggs Hatch, Activities of Acetyl-CoA Carboxylase and Fatty Acid Synthase of Yolk-Sac Larvae of Hybrid Grouper (Epinephelus fuscoguttatus ♀ × Epinephelus polyphekadion ♂)
In order to investigate the effects of glucose on fertilized eggs hatch,and activities of acetyl-CoA carboxylase (ACC) and fatty acid synthase (FAS) of yolk-sac larvae of hybrid grouper,different concentrations of glucose were added to the water for hatching of hybrid grouper (Epinephelus fuscoguttatus ♀ ×Epinephelus polyphekadion ♂),which were 0,2,4,6,8 and 10 mg/L,respectively.The hatching rate and abnormality rate were observed,and the total length,activities of ACC and FAS of yolk-sac larvae at 1,2 and 3 days of age were tested.The results showed as follows:hatching rate of fertilized eggs and abnormalities of newly hatched larvae were significantly affected by glucose concentration (P<0.05);the correlation between the concentration of glucose and hatching rate of fertilized eggs and abnormality rate of newly hatched larvae both could be described by quadratic equations.The optimum glucose concentration was 3.8 mg/L to reach the highest hatching rate of fertilized eggs.When the concentration of glucose was below 6 mg/L,hatching rate of fertilized eggs increased,while abnormality rate of newly hatched larvae decreased,total length of yolk-sac larvae increased with the increase of concentration of glucose.For yolk-sac larvae at 1,2 and 3 days of age,activities of ACC and FAS in glucose supplemented group were significantly higher than those in control group (P<0.05).With the increase of concentration of glucose,the activities of ACC and FAS of larvae at 1 day of age increased;at 2 and 3 days of age,ACC activity of larvae displayed a'up-down'trend and FAS activity showed a'rising-gentle'trend.It is concluded that in artificial breeding of hybrid grouper,the supplementation of 6 mg/L glucose in hatching water can promote fertilized egg hatch and yolk-sac larvae development,and significantly increase ACC and FAS activities of yolk-sac larvae.
