Optimization of Fermentation Process for Cellulase Production by Bacillus mojavensis HXS-LV12 Using Plackett-Burman and Box-Behnken Experiments
In this study,the fermentation condition of cellulase production by Bacillus amyloliquefaciens HXS-LV12 was optimized to produce cellulase.Firstly,cultural conditions(variety of carbon sources,mass concen-tration of carbon source,variety of nitrogen sources,mass concentration of nitrogen sources,variety of inor-ganic salts,mass concentration of inorganic salt,rotate speed,temperature,inoculum volume,initial pH and fermentation time)of HXS-LV12 were optimized by single-factor experiments.Then,three factors,signally affect production of cellulase by HXS-LV12 fermentation,were obtained by Plackett-Burman experiment.The maximum response area of cellulase activity was determined by steepest ascent.Finally,Box-Behnken experi-ment were used to ascertain the optimal condition of cellulase production.The optimal conditions were:sodium carboxymethylcellulose(CMC-Na)mass concentration was 32.9 g/L,yeast extract powder mass concentration was 17.6 g/L,K2HPO4 was 5.7 g/L,rotate speed was 190 r/min,inoculum volume was 6.5%,fermentation time was 24 h,initial pH was 7.0 and incubating temperature was 33℃.Under the optimal conditions,the ac-tivity of cellulase produced by HXS-LV12 were achieved(40.53±0.43)U/mL,which was 2.25 times of that before optimization.[Chinese Journal of Animal Nutrition,2024,36(1):564-578]
