饲粮中添加不同水平双歧杆菌对脂多糖应激雏鹅回肠损伤的缓解效果

本试验旨在研究饲粮中添加不同水平双歧杆菌对脂多糖(LPS)应激雏鹅回肠损伤的缓解效果.选取288只1日龄江南白鹅公雏,随机分为6组,每组6个重复,每个重复8只.采用3×2两因素设计,两因素分别为双歧杆菌添加水平(分别添加0、300和600 mg/kg双歧杆菌冻干粉,有效活菌数为1.7×1010 CFU/g)和LPS应激与否(腹腔注射LPS生理盐水溶液或生理盐水,LPS生理盐水溶液浓度为0.5 mg/mL).试验期共28 d,分别于试验第15、17和19天对雏鹅进行1次腹腔注射,注射量均为1 mL/kg BW.结果表明:1)LPS应激显著提高了雏鹅血清中二胺氧化酶(DAO)活性和D-乳酸(D-LA)含量(P<0.05);饲粮中添加双歧杆菌显著降低了雏鹅血清中DAO活性和D-LA含量(P<0.05).2)LPS应激显著提高了雏鹅回肠损伤评分(P<0.05),并对回肠黏蛋白2(MUC2)含量产生显著影响(P<0.05);饲粮中添加双歧杆菌显著降低了雏鹅回肠损伤评分(P<0.05),且显著提高了回肠MUC2含量(P<0.05).3)与饲粮中添加300 mg/kg双歧杆菌相比,饲粮中添加双歧杆菌600 mg/kg双歧杆菌除28日龄时显著降低雏鹅血清中D-LA含量且显著提高回肠MUC2含量(P<0.05)外,其余日龄及指标均无显著差异(P>0.05).4)LPS应激会造成雏鹅回肠上皮细胞严重损伤,胞质水肿明显,细胞核近似不规则椭圆形,高尔基体囊膜扩张,微绒毛出现整体结构损伤,细胞间紧密连接及中间连接结构模糊.饲粮中添加双歧杆菌通过诱导细胞自噬参与缓解上述现象,并且有助于保护线粒体结构,尤其以添加300 mg/kg双歧杆菌冻干粉可以明显缓解胞质水肿和粗面内质网扩张.综上所述,1~28日龄雏鹅LPS应激会对回肠及回肠上皮细胞造成损伤,致使MUC2分泌异常并增大肠道通透性.饲粮中添加双歧杆菌能通过促进MUC2分泌缓解回肠损伤并改善肠道通透性,并通过诱导细胞自噬参与缓解回肠上皮细胞损伤.在本试验条件下,饲粮中添加600 mg/kg双歧杆菌冻干粉未能取得更好的缓解效果,因此建议添加300 mg/kg.
Alleviating Effects of Different Dietary Levels of Bifidobacterium on Ileum Injury of Goslings under Lipopolysaccharide Stress
The aim of this study was to investigate the alleviating effects of different dietary levels of Bifidobac-terium on ileum injury of goslings under lipopolysaccharide(LPS)stress.A total of 288 one-day-old male Jiangnan white goose chicks were randomly divided into 6 groups with 6 replicates per group and 8 chicks per replicate.A 3×2 two-factor design was adopted,and the two factors were Bifidobacterium supplemental level(0,300 and 600 mg/kg Bifidobacterium freeze-dried powder was supplemented,and the effective viable bac-teria number was 1.7×1010 CFU/g,respectively)and LPS stress or not(intraperitoneal injection of LPS saline solution or saline solution,and the concentration of LPS saline solution was 0.5 mg/mL).The experiment las-ted for 28 days,and the goslings were intraperitoneally injected with 1 mL/kg BW on the 15th,17th and 19th days of the experiment,respectively.The results showed as follows:1)LPS stress significantly increased the diamine oxidase(DAO)activity and the D-lactic acid(D-LA)content in serum of goslings(P<0.05);Bifidobacterium supplementation significantly reduced the DAO activity and the D-LA content in serum of gos-lings(P<0.05).2)LPS stress significantly increased the ileum injury score of goslings and had a significant effect on mucin 2(MUC2)content in ileum(P<0.05);Bifidobacterium supplementation significantly re-duced the ileum injury score and increased the MUC2 content in ileum(P<0.05).3)Compared with the sup-plementation of 300 mg/kg Bifidobacterium,the supplementation of 600 mg/kg Bifidobacterium significantly decreased the D-LA content in serum and significantly increased the MUC2 content in ileum of goslings at 28 days of age(P<0.05),but there were no significant differences in the indices at other days of age(P>0.05).4)LPS stress could cause serious injury of gosling's ileal epithelial cells,with obvious cytoplasm edema,ap-proximately irregular oval nucleus,expansion of Golgi capsule membrane,overall structural damage of mi-crovilli,and fuzzy structure of tight junctions and intermediate junctions between cells.Bifidobacterium was in-volved in alleviating the above phenomenon by inducing autophagy and helping to protect mitochondrial struc-ture.In addition,300 mg/kg Bifidobacterium freeze-dried powder could significantly alleviate cytosolic edema and rough endoplasmic reticulum dilation.In conclusion,LPS stress of goslings aged from 1 to 28 days can cause damage to the ileum and ileal epithelial cells,result in abnormal secretion of MUC2 and increase intesti-nal permeability.Dietary Bifidobacterium can alleviate ileum injury and improve intestinal permeability by pro-moting MUC2 secretion,and can alleviate ileal epithelial cell injury by inducing autophagy.Under the condi-tions of this experiment,the supplementation of 600 mg/kg Bifidobacterium freeze-dried powder in the diet could not achieve better relief effects,so the supplementation of 300 mg/kg is recommended.[Chinese Jour-nal of Animal Nutrition,2024,36(5):3035-3048]
