Anti-Inflammatory Activity and Mechanism of Total Flavonoids from Veronica linariifolia on Mouse Macrophage
The purpose of this study was to investigate the in vitro anti-inflammatory activity and mechanism of total flavonoids from Veronica linariifolia by lipopolysaccharide(LPS)-induced mouse macrophages RAW264.7.MTT assay was used to detect the cytotoxicity of total flavonoids from Veronica linariifolia;the phagocytosis capacity was determined by neutral red assay;the production of nitric oxide(NO)was detected by Griess method;enzyme-linked immunosorbent assay(ELISA)method was used to detect the production of interleukin-1β(IL-1β),interleukin-6(IL-6),tumor necrosis factor-α(TNF-α),prostaglandin E2(PGE2),inducible nitric oxide synthase(iNOS)and cyclooxygenase-2(COX-2);the expression levels of nuclear fac-tor-κB(NF-κB)signaling pathway-associated proteins were assayed by Western blotting method.The results showed that the total flavonoids from Veronica linariifolia in the mass concentration of 200 μg/mL had no sig-nificant cytotoxicity to mouse macrophages RAW264.7(P>0.05).Different mass concentrations(25,50,100,200,300 and 400 μg/mL)of total flavonoids from Veronica linariifolia could significantly or extremely significantly enhance the phagocytosis capacity of mouse macrophages RAW264.7 induced by LPS(P<0.05 or P<0.01),significantly or extremely significantly inhibit the production of NO,IL-1β,IL-6,TNF-α,PGE2,iNOS,COX-2 and the protein expression levels of phosphorylated nuclear factor-KB p65(p-NF-KB p65)and phosphorylated inhibitor of nuclear factor-κBα(p-IκBα)(P<0.05 or P<0.01),and significantly or extremely significantly enhance the protein expression level of inhibitor α of nuclear factor-κB(IκBα).In conclusion,the total flavonoids from Veronica linariifolia can enhance the phagocytosis function of mouse macrophages and exert anti-inflammatory activity in vitro by inhibiting the production of iNOS,COX-2,NO and inflammatory factors,and regulating the expression of NF-κB signaling pathway-associated proteins.[Chinese Journal of Animal Nutrition,2024,36(6):3963-3972]
