Immunoadjuvant Effects of Allium mongolicum Regel Polysaccharide on Ovalbumin in Mice
The aim of this experiment was to study the adjuvant effects of Allium mongolicum Regel polysac-charide(AMRP)on mice immunized with model antigen ovalbumin(OVA).Forty specific pathogen free(SPF)female Kunming mice aged 6 to 8 weeks and weighing(20±2)g were randomly divided into 5 groups,which were blank control group(BC group),OVA immune control group(OVA group),OVA+low-dose AMRP group(LAMRP group),OVA+medium-dose AMRP group(MAMRP group)and OVA+high-dose AMRP group(HAMRP group),with 8 replicates per group and 1 replicate per replicate.The pre-trial period lasted for 7 days and the experimental period lasted for 34 days.During days 1 to 4 and days 16 to 19,the mice in BC group and OVA group were administrated with normal saline by oral gavage,and those in LAMRP group,MAMRP group and HAMRP group were administrated with 15,20 and 25 mg/kg BW AMRP by oral gavage,respectively,with an amount of 0.2 mL.On day 5 and day 20,the mice in BC group were injected subcutaneously with 0.1 mL normal saline every mouse,and those in the other groups were injected with 100 μg OVA solution every mouse.Samples were collected for 2 weeks after second immunization.The results showed as follows:1)compared with BC group and OVA group,different doses of AMRP had no significant effects on body weight and weight gain rate of mice at each stage of the experiment(P>0.05).2)Compared with BC group and OVA group,there were no significant differences in thymus index of mice in each dose of AMRP group(P>0.05);the spleen index(P<0.01)and germinal center diameter in spleen(P<0.05)in MAMRP group were significantly increased.3)Compared with BC group and OVA group,the serum interleu-kin-10(IL-10)content of mice in MAMRP group and HAMRP group was extremely significantly increased(P<0.01);compared with OVA group,the serum interferon-γ(IFN-γ)content in LAMRP group and MAM-RP group was significantly increased(P<0.05);there were no significant differences in serum contents of in-terleukin-2(IL-2)and interleukin-4(IL-4)among all groups(P>0.05).4)Compared with BC group,the OVA specific antibody immunoglobulin G(IgG)content in serum of mice in MAMRP group was extremely significantly increased(P<0.01).In conclusion,AMRP can improve the immune ability of mice against anti-gen OVA,and then show certain adjuvant effects,which can be used as an effective vaccine adjuvant to induce immune response;under the experimental conditions,the adjuvant effects are better when 20 mg/kg BW AM-RP is administrated by oral gavage.[Chinese Journal of Animal Nutrition,2024,36(9):6007-6014]
