美洲大蠊抗菌肽的提取及其对不同致病菌抑菌活性的影响

本试验旨在探究美洲大蠊抗菌肽的最佳提取时间及其对不同致病菌抑菌活性的影响.选取100只5龄美洲大蠊,随机分为2组,每组5个重复,每个重复10只.试验以大肠杆菌为诱导源诱导美洲大蠊产生抗菌肽,诱导组注射1 μL 1×108 CFU/kg的大肠杆菌,对照组注射1 μL的生理盐水,分别在诱导后12、24、36、48、60和72 h提取美洲大蠊抗菌肽.通过十二烷基硫酸钠-聚丙烯酰胺凝胶电泳(SDS-PAGE)试验确定美洲大蠊抗菌肽的分子质量,通过BCA试剂盒测定美洲大蠊抗菌肽提取液的蛋白浓度,通过药敏片法测定美洲大蠊抗菌肽提取液的抑菌活性,同时测定其最小抑菌浓度(MIC)、热稳定性、pH稳定性和反复冻融稳定性.结果表明:1)美洲大蠊抗菌肽的分子质量在7.5 ku左右.诱导后12、24、36、48、60、72 h,诱导组美洲大蠊抗菌肽提取液的蛋白浓度极显著高于对照组(P<0.01),诱导后48 h时美洲大蠊抗菌肽提取液的蛋白浓度最高,因此选择48 h时诱导产生的美洲大蠊抗菌肽进行后续的抑菌试验.2)与对照组相比,诱导组的大肠杆菌、沙门氏菌和金黄色葡萄球菌的抑菌圈直径极显著升高(P<0.01).美洲大蠊抗菌肽对大肠杆菌的最小抑菌浓度为1.0 mg/L,对沙门氏菌的最小抑菌浓度为3.0 mg/L,对金黄色葡萄球菌的最小抑菌浓度为2.5 mg/L.3)与对照组相比,经75、85、95、100 ℃水浴加热后的美洲大蠊抗菌肽提取液的抑菌直径显著降低(P<0.05),冻融2、4、6、8、10次的美洲大蠊抗菌肽提取液的抑菌圈直径显著降低(P<0.05),pH为1、3、7、9、11溶液处理后的美洲大蠊抗菌肽提取液的抑菌圈直径显著降低(P<0.05).综上所述,大肠杆菌诱导后48 h为美洲大蠊抗菌肽的最佳提取时间,同时具备浓度高、热稳定性强和耐酸碱等特性.
Extraction of Antimicrobial Peptides from Periplaneta americana and Their Effects on Antibacterial Activity of Different Pathogenic Bacteria
This experiment was conducted to explore the optimal extraction time of antimicrobial Peptides from Periplaneta americana and their effects on antibacterial activity of different pathogenic bacteria.A total of 100 Periplaneta americana were randomly divided into 2 groups with 5 replicates per group and 10 Periplaneta a-mericana per replicate.The experiment used Escherichia coli as the inducer to induce the production of antimi-crobial peptides from Periplaneta americana,the induction group was injected with 1 µL 1×108 CFU/kg Esch-erichia coli,while the control group was injected with 1 μL physiological saline,the antimicrobial peptides from Periplaneta americana were extracted at 12,24,36,48,60 and 72 h after induction,respectively.The molecular weight of antimicrobial peptides from Periplaneta americana was determined by sodium dodecyl sul-fate-polyacrylamide gel electrophoresis(SDS-PAGE)test,the protein concentration of extracting solution of antimicrobial peptides from Periplaneta americana was determined by BCA kit,the antibacterial activity of ex-tracting solution of antimicrobial peptides from Periplaneta americana was determined by the drug sensitive tab-let method,and the minimum inhibitory concentration(MIC),thermal stability,pH stability and repeated freeze-thaw stability were also determined.The results showed as follows:1)the molecular weight of the anti-microbial peptides from Periplaneta americana was about 7.5 ku.After induction at 12,24,36,48,60 and 72 h,the protein concentration of extracting solution of antimicrobial peptides from Periplaneta americana of induction group was significantly higher than that of the control group(P<0.01),the protein concentration of extracting solution of antimicrobial peptides from Periplaneta americana after induction at 48 h was the high-est,therefore,selected the antimicrobial peptides from Periplaneta americana after induction at 48 h for the subsequent antibacterial tests.2)Compared with the control group,the diameter of antibacterial circle of Esch-erichia coli,Salmonella and Staphylococcus aureus of induction group was significantly increased(P<0.01).The antimicrobial peptides from Periplaneta americana minimum inhibitory concentration against Escherichia coli was 1.0 mg/L,the minimum inhibitory concentration against Salmonella was 3.0 mg/L,and the minimum inhibitory concentration against Staphylococcus aureus was 2.5 mg/L.3)Compared with the control group,the diameter of antibacterial circle of extracting solution of antimicrobial peptides from Periplaneta americana after heating in water baths at 75,85,95 and 100 ℃ was significantly decreased(P<0.05),the diameter of antibacterial circle of extracting solution of antimicrobial peptides from Periplaneta americana after freeze-thaw 2,4,6,8 and 10 times was significantly decreased(P<0.05),and the diameter of antibacterial circle of ex-tracting solution of antimicrobial peptides from Periplaneta americana after treatment with solutions with pH of 1,3,7,9 and 11 was significantly decreased(P<0.05).In conclusion,the 48 h after Escherichia coli induc-tion is the optimal extraction time for antimicrobial peptides from Periplaneta americana,which has the charac-teristics of high concentration,strong thermal stability and acid and alkali resistance.[Chinese Journal of Ani-mal Nutrition,2024,36(9):6072-6082]
