Protective Effect of Raffinose on Liver Oxidative Injury Induced by Aflatoxins B1 in Mice
The aim of this study was to investigate the protective effect of raffinose(Raf)on the liver oxida-tive damage induced by aflatoxins B1(AFB1)in mice.Twenty C57BL/6 male mice aged 6 to 8 weeks were randomly divided into 5 groups,they were control group,Raf group(50 mg/kg BW Raf was given),AFB1 group(1.5 mg/kg BW AFB,was given),low dose RAF treatment group(25 mg/kg BW Raf and 1.5 mg/kg BW AFB1 were given),high dose Raf treatment group(50 mg/kg BW Raf and 1.5 mg/kg BW AFB1 were given),each group had 4 mice.Mice in each group were given a gavage per two days within 30 d.After gavage period,the growth performance,biochemical indexes,expression of related proteins were meas-ured and the tissue sections were observed.The results showed as follows:1)after Raf treatment,the feed in-take of AFB1-induced mice was reduced,and the decreasing trend of weight gain rate was inhibited.2)Raf could inhibit the extremely significantly increase of serum aspartate aminotransferase(AST)and alanine amin-otransferase(ALT)activities induced by AFB1(P<0.01).3)Pathological observation of liver tissue showed that,compared with the control group,there were more variable cells in AFB,group with uneven arrange-ment,which indicated liver cell damage,and that was improved after Raf took effect.4)Raf could inhibit the extremely significantly decrease of liver apoptosis-related protein B-cell lymphoma-2(Bcl-2)protein expres-sion level(P<0.01)and the extremely significantly increase of Bcl-2-associated X protein(Bax)and cyste-ine-aspartic acid protease-3(Caspase-3)expression levels induced by AFB1(P<0.01).5)RAF could signifi-cantly decrease the content of malondialdehyde(MDA)in liver of AFB1-induced mice(P<0.01),and signif-icantly increase the activity of superoxide dismutase(SOD)and the content of reduced glutathione(GSH)in liver of AFB1-induced mice(P<0.01).6)RAF could inhibit the extremely significantly increase of liver Kelch-like ECH-related protein 1(Keap1)protein expression level(P<0.01)and the extremely significantly decrease of heme oxygenase-1(HO-1)in liver of mice induced by AFB1(P<0.01).In conclusion,Raf plays a role in improving weight gain rate decrease,liver function damage and liver oxidative injury in mice induced by AFB1.[Chinese Journal of Animal Nutrition,2024,36(12):8100-8109]
