Optimization Test of Rooting Agent for Castanopsis hystrix Cutting
The super seedlings of Castanopsis hystrix were used as the cutting mother plant,and the levels of endogenous hor-mones indole acetic acid(IAA),abscisic acid(ABA),gibberellin(GAs)and zeatin(ZR)in the cuttings during the rooting process of C.hystrix cuttings as well as changes in the effect of cutting seedlings under the treatment of 16 growth regulators were analyzed.The results showed as follows:during the rooting process of 0-60 d cutting,with the extension of cutting time,the level of IAA increased first and then decreased,and the level of GAs decreased first and then increased;the level of ABA increased at the 15 d cutting,and then decreased and remained at a relatively stable level at 30-60 d cutting,and there was no significant change in ZR.The rooting rates were significantly different(0%-94.8%,P<0.05)under 16 rooting a-gents of different ratios composed of exogenous auxin substances 0-400 mg·L-1 IAA,naphthalene acetic acid(NAA),indole butyric acid(IBA)and 0-200 mg·L-1 Gas,synthesis inhibitors paclobutrazol(PBZ)and paclobutrazol(DPC).Through the range size(R)analysis,it was found that DPC(32.3)>NAA(28.5)>IBA(25.6)>PBZ(23.5)>IAA(10.9).There was no significant correlation between cutting survival rate and rooting rate(R2=0.51,P>0.05),but there was a signifi-cant positive correlation linear relationship between cutting survival rate and root number(R2=0.91,P<0.05).The lignifi-cation degree of cuttings affected the effect of cutting seedling.Compared with the initial lignification and lignification cuttings,the amplitude of fluctuation of the rooting rate,root number and survival rate of semi-primed cuttings was smaller,and the cut-ting seedling efficiency was the highest(95.4%)under the treatment of 200 mg·L-1 NAA+200 mg L-1 IBA+100 mg· L-1 PBZ+100 mg·L-1 DPC.It is considered that using semi-lignified spikes as cuttings and 200mg·L-1 NAA+200 mg ·L-1 IBA+100 mg·L-1 PBZ+100 mg·L-1 DPC as rooting agent could achieve better cutting effect.
