超高效液相色谱-串联质谱法测定人血中的依托咪酯

本研究建立了超高效液相色谱-质谱(UPLC/MS/MS)检验全血中依托咪酯的方法.经过乙腈沉淀蛋白提取净化,应用电喷雾离子源(ESI)、多反应监测(MRM)正离子模式质谱进行定性及定量检验血样中依托咪酯.全血中依托咪酯在0.25~20 ng/mL范围内呈现良好的线性关系(R2=0.999),方法最低检出限为0.05 ng/mL,定量限为0.25 ng/mL.在0.25、1.0、10 ng/mL加标水平下,方法回收率为93.7%~98.5%,相对标准偏差均小于6.3%.该方法样品前处理操作简便,准确度好,灵敏度高,适用于全血中依托咪酯的定性定量分析.
Determination of Etomidate in Blood Samples by UPLC-MS/MS
In this study,a method for the determination of etomidate in blood samples by UPLC-MS/MS was established.The whole blood were extracted by acetonitrile,then etomidate was determined with the UPLC-MS/MS system coupled with ESI+mode and multiple reaction monitoring(MRM).Etomidate gave linear calibration curves in the range of 0.25-20 ng/mL with correlation coefficients(R2)better than 0.999.The detection limit(LOD)of etomidate was 0.05 ng/mL and the quantification limit(LOQ)was 0.25 ng/mL.At spiked levels of 0.25、1.0、10 ng/mL,the recoveries were in the range of 93.7%-98.5%with relative standard deviations(RSDs)less than 6.3%.The method is simple,rapid,sensitive and accu-rate for the determination of trace etomidate in whole blood samples.
