公石松多糖的提取、结构表征及生物活性研究

以公石松为原料,通过水提醇沉法获得公石松多糖.利用高效凝胶渗透色谱、柱前衍生化HPLC法、紫外、红外光谱、刚果红实验等对公石松多糖的结构进行了表征,通过抗氧化实验评估公石松多糖的体外抗氧化活性,采用MTT法初步探讨了其对肝癌细胞HepG2、肝细胞LO2的抑制效果.结果表明,公石松多糖的相对分子量分布于50 325~115 144 Da,由甘露糖、葡萄糖、半乳糖和阿拉伯糖组成,具有三螺旋结构.体外抗氧化活性测定显示,公石松多糖对DPPH、ABTS、羟基自由基表现出较强的清除能力.抗肿瘤实验表明,公石松多糖具有抑制HepG2增殖的能力.
Extraction,structural characterization and bioactivity of polysaccharides from Gongshisong
Gongshisong polysaccharide was obtained from Gongshisong by water extract-alcohol precipitation method.The structure of Gongshisong polysaccharide was characterized by high performance gel permeation chromatography,pre-col-umn derivatization HPLC,ultraviolet and infrared spectroscopy,and Congo red assay,etc.The antioxidant activity of Gong-shisong polysaccharide was evaluated by antioxidant assay,and the inhibitory effect of Gongshisong polysaccharide on hepa-tocellular carcinoma cells HepG2 and hepatocellular cells LO2 was preliminarily investigated by MTT assay.The results showed that the relative molecular weight of Gongshisong polysaccharide was distributed from 50,325 to 115,144 Da,and consisted of mannose,glucose,galactose and arabinose with triple-helical structure.In vitro antioxidant activity assay showed that Gongshisong polysaccharide exhibited strong scavenging ability against DPPH,ABTS,and hydroxyl radicals.Anti-tumor assay showed that Gongshisong polysaccharide has the ability to inhibit the proliferation of HepG2.
