[目的]为培养优质的桑黄菌,筛选适宜的母种培养基配方.[方法]将桑黄菌种接种到不同配方培养基进行初筛、复筛,通过形态比较,从菌丝长势及长速等方面综合考虑,选出最优母种培养基配方.[结果]初筛结果表明,选择桑黄菌丝生长快且浓密、不易老化、不吐黄水的蔗糖和葡萄糖为较佳碳源;选择生长快且不易老化、长势好、不吐黄水的蛋白胨和胰蛋白胨作为较佳氮源.复筛结果表明,以1/4葡萄糖+3/4蔗糖为配方的母种培养基,桑黄菌丝的长势最好、浓密、长速较快.[结论]生产上推荐桑黄菌母种培养基采用配方为1/4葡萄糖+3/4蔗糖,即马铃薯200 g、葡萄糖5 g、蔗糖15 g、琼脂20 g、水1 000 mL.
Study on Formula Optimization of Mother Culture Medium of Sanghuangporus
[Objective]To screen the best mother medium of Sanghuang for cultivating high-quality Sanghuangporus.[Method]Sanghuangporus strain was inoculated into the medium with different formulas for preliminary screening and re-screening.Through morphological comparison,the optimal mother culture medium formula was selected from the aspects of hypha growth vigor,growth rate and others.[Result]The results of preliminary screening showed that sucrose and glucose were selected as the best carbon sources with fast growth,not easy to aging,no yellow water,and dense growth.Peptone and tryptone were selected as the best nitrogen source with fast growth,not easy to aging,good growth vigor and no yellow water.The results of re-screening showed that the hypha had good growth vigor,dense and fast growth when formula of 1/4 glucose+3/4 sucrose was used.[Conclusion]The formula of 1/4 glucose+3/4 sucrose is recommended in production,containing 200 g potato,5 g glucose,15 g sucrose,20 g agar and 1 000 mL water.
