大球盖菇颗粒原种培养基筛选及其应用

[目的]为筛选适宜的大球盖菇原种培养基,研发菌丝活力强、萌发点多、松散均一的颗粒原种,并确立合适的扩繁透气袋栽培接种量.[方法]以'大球盖菇 8 号'为试验菌株,采用菌丝生长速度为评价指标,通过单因素试验,比较由小米、细木屑、玉米芯、菜籽粕等主要组分的不同培养基的颗粒原种扩繁栽培种效果,对比不同接种量对透气袋栽培种满袋时间的影响.[结果]确定最佳大球盖菇颗粒原种培养基配方为XGM(小米 50%、蔗糖 2%、细木屑 20%、硅藻岩 26%、轻质碳酸钙2%),培养基的碳氮比是 32.5,培养的大球盖菇颗粒原种菌丝生长速度为 2.99 mm/d.颗粒原种XGM扩繁透气袋栽培种最佳接种量为 1.25%,透气袋栽培种平均满袋时间为 13.4 d.[结论]通过研究优化了颗粒原种培养基,初步建立透气袋栽培种生产工艺,为工厂化生产大球盖菇透气袋栽培种奠定基础.
Study on Selection and Application of Culture Medium of Particle Pre-culture Spawn of Stropharia rugosoannulata
[Objective]The aim is to screen suitable spawn culture media for Stropharia rugosoannulata and develop particle pre-culture spawn with strong mycelial vitality,numerous germination points,and loose uniformity.The appropriate inoculation amount of spawn using vent plastic bag was established.[Method]Using'S.rugosoannulata No.8'as the experimental strain and the mycelial growth rate as the evaluation index,a single factor experiment was conducted to compare the effects of particle spawn propagation and cultivation on different media containing main components such as millet,fine sawdust,corn cob,and rapeseed meal,and compare the effects of different inoculation amount on the bag-filled time of spawn using vent plastic bag.[Result]The optimal formula for the particle pre-culture spawn medium of S.rugosoannulata was determined to be XGM,with 50%millet,2%sucrose,20%fine sawdust,26%diatomaceous earth,and 2%light calcium carbonate.The carbon-to-nitrogen ratio in the medium was 32.5,and the growth rate of the mycelia of the particle pre-culture spawn was 2.99 mm/d.The optimal inoculation amount was 1.25%and the average bag-filled time was 13.4 days using particle pre-culture spawn XGM to expand spawn in vent plastic bag.[Conclusion]The culture medium of particle pre-culture spawn was optimized and the production process of S.rugosoannulatain using vent plastic bag was preliminarily established,which laid a foundation for industrial production of S.rugosoannulata spawn.
