Influence of Different Preservation Conditions on the Growth of Volvariella volvacea Hyphae
[Objective]To explore the environmental conditions suitable for the preservation of Volvariella volvacea strain for 10 months and ensure the excellent characteristics of the existing V.volvacea strains.[Method]Taking V.volvacea'V9715'as the test strain,three preservation condition factors such as preservation temperature,preservation substrate,and additive were set.The preservation effect was analyzed and evaluated from aspects such as the germination and growth of hyphae,the number of chlamydospores,and enzyme activity to explore the influence of different preservation factors on the preservation effect of V.volvacea strains after 10 months of preservation.[Result]After 10 months of preservation under different preservation conditions,there were differences in the germination and growth of V.volvacea hyphae,the formation time of chlamydospores,and enzyme activity.Among them,the germination rate of hyphae under preservation condition treatment 3(20℃+corncob+liquid paraffin)was the highest at 88.30%,the germination days were 2 days,the formation time of chlamydospores is 11.5 days,and growth days to fill petri dish were the shortest at 4 days.The hyphal growth rate was the fastest at 1.14 cm/d.The hyphae have strong growth potential,high density of aerial hyphae,and medium mycelial biomass while the mycelial biomass was the largest(0.36 g)under preservation condition treatment 1(8℃+cottonseed hull+no additive).There is no significant difference among other treatments.The activities of cytochrome P450 oxidoreductase(CPR)and polyphenol oxidase(PPO)of hyphae under treatment 3(20℃+corncob+liquid paraffin)are the lowest,which were 2.16 mg/g FW and 36.91 U/g FW,respectively.The activity of total amylase(AMY)was the highest,which was 11.88 mg/(min·g)FW.[Conclusion]Under the preservation condition of treatment 3(20℃+corncob+liquid paraffin),the preservation effect of V.volvacea strains for 10 months was the best.
