低磷胁迫下杉木针叶蛋白的提取与表达差异分析

采用TCA-丙酮沉淀高速离心法,对杉木针叶蛋白质组双向电泳技术中全细胞蛋白的提取、蛋白的裂解、电泳条件等环节进行了优化改良,获得了高重复性、高清晰度的杉木针叶响应低磷胁迫的差异蛋白图谱.在此基础上对低磷处理5d的杉木进行针叶蛋白提取,并使用ImageMaster 2D Platinum 7.0软件分析比较杉木针叶在对照与低磷处理下的蛋白表达谱.研究表明,图谱中有59个蛋白点存在显著差异,可能与低磷胁迫有关,其中,上调表达点52个,下调表达点7个.
Variance analysis of protein extraction and expression for Chinese fir needles under low phosphorus stress
In order to obtain the protein profile with high repeatability and resolution under the condition of phosphorus deficiency,the high-speed centrifugation of TCA-acetone precipitation and the optimized electrophoresis including the extraction of whole-cell protein in Chinese fir needles,the protein cleavage and the condition of electrophoresis were used in this study.The protein extraction of fir needles under 5 days low-phosphorus treatment was conducted,and the comparison of protein profile between control and the treatment samples were made by using Image Master 2D Platinum 7 software.The results showed that there were 59 protein points of low phosphorus treatment significantly different from the control sample,which included 52 upregulated and 7 downregulated points.
