高产纤维素酶菌株的鉴定及其活性特征

Identification of strains producing cellulase and its enzyme activity determination

为分析倒木中具有高产纤维素酶菌株的羧甲基纤维素(CMC)酶活性的大小,以天宝岩国家级自然保护区内长苞铁杉倒木作为研究对象,将菌源通过富集培养、滤纸崩解、CMC酶活测定,分离筛选出具有高效降解纤维素的菌株,并将筛选出的高产纤维素酶菌株进行18S rRNA测序分析鉴定.结果表明,经过初筛、复筛,得出TB4为高产纤维素酶的目的菌株,经鉴定该菌株为小刺青霉,该菌株产纤维素酶的酶活性为243.73 U·mL-1.
Based on cumulation cultivation,filter paper disintegrating,carboxymethyl cellulose (CMC) enzyme activity determination,strains with great capacity in cellulose decomposition were isolated to analyze the CMC enzyme activities of fallen logs in the Tsuga longibracteata W.C.Cheng forest of Tianbaoyan National Nature Reserve.18S rRNA sequencing analysis of the selected stains was carried out and identified as Penicillium spinulosum which the CMC enzyme activity was 243.73 U·mL-1.The P.spinulosum strain had the ability in cellulose degradation in the T.longibracteata forest of Tianbaoyan National Nature Reserve.
