In this paper, 10 primer combinations screened from 256 of sequence-related amplified polymorphism ( SRAP ) were used to analyze genetic diversity of 17 cultivars of Castanea henryi which grown in Jian′ou City of Fujian Province. The results showed high genetic diversity in these cultivars since 10 primer combinations obtained 200 bands containing the 183 polymorphic bands which took 91.5% proportion. The primer combinations em2me8 and em5me9 showed 100% polymorphism rate. The genetic similarity coefficient was between 0. 5082 and 0. 7978, and the average was 0. 6530, DJZ and ZJZ have the closest genetic relationship while DJZ and SFZ have the farthest genetic relationship. As the dendrogram by SRAP marker, at the genetic similarity coefficient of 0.6557, 17 C. henryi cultivars were divided into 3 classes:the first class had 3 cultivars ( CSH, SFZ and WKCM) , the second class contained 11 cultivars (DBZ, NJZWS, WZ, TZ, NJZZS, ONZ, HZZ, HZ, YTZ, BLZ and YZ), and the rest 3 cultivars (DJZ, ZJZ and XJZ) belonged to the last class. A SRAP fingerprint was built for the 17 C. henryi cultivars by em2me8. The paper provides a basis for selecting parents of crossbreeding and genome mapping.
