摘要

采用组织分离法从福建省5个地区的桉树叶片中获得87株内生真菌,鉴定为7目及2个未定目,含15属及1个未定属,20种和1个未定种;其中Pseudoplagiostoma eucalypti菌株数量占总数的0.32,为优势种类.龙岩采样点桉树叶片内生真菌辛普森多样性指数为0.92,福州采样点的桉树内生菌香农-威纳指数为2.13,分别高于其它4个地点.经筛选,发现87株内生真菌中,哈茨木霉(Trichoderma harzianum)ND-13的菌丝能够覆盖桉树焦枯病菌(Calonectria pseudoreteaudii)菌落并抑制其生长;显微观察显示,ND-13菌丝缠绕病原菌菌丝;检测发现ND-13不产生几丁质酶,但产生β-葡聚糖酶.研究结果显示桉树叶片内生真菌种类丰富;ND-13具有防治桉树焦枯病的潜力,表明从桉树内生真菌中筛选抗桉树焦枯病菌是可行的.

Abstract

By tissue separation method, 87 endophytic fungal strains were isolated from Eucalyptus leaves which were collected from 5 sites in Fujian Province.All the endophytic strains were identified into 7 orders and 2 unidentified orders, 15 genera and 1 unidentified genus, 20 species and 1 unidentified species.Pseudoplagiostoma eucalypti strains were the dominant species accounting for 0.32 of the total.The Simpson diversity index of Eucalyptus endophytic fungi was 0.92 and Shannon-Wiener index of endophytic fungi isolated from Fuzhou was 2.13 which were the highest in the 5 sites, respectively.By dual-culture test, only Trichoderma harzianum ND-13 exhibited antifungal activity against Calonectria pseudoreteaudii.Colony of C.pseudoreteaudiiwas covered with T. harzianum ND-13 in dual-culture.Microscopic observation showed that ND-13 mycelia could enwind C.pseudoreteaudii hyphae.The study found that ND-13 did not produce chitinase, but β-glucanase.The results showed that the diversity of endophytic fungi in Eucalyptus leaves was high, and ND-13 had the potential to control the Calonectria leaf blight ( CLB) of Eu calyptus spp.;and it was feasible to obtain biocontrol agents of CLB from Eucalyptus endophytic fungi.
