Effects of long non-coding RNA AC004920.3 on cancer embolus and cells of gastric adenocarcinoma
Objective To investigate the expression of AC004920.3 in serum of gastric adenocarcinoma patients and its effect on cells.Methods Serum samples and medical records were collected.The expression of AC004920.3 was amplified by PCR.After transfecting AGS cells with lentivirus overexpression of AC004920.3,apoptosis,CCK proliferation and scratches were performed.Results Serum AC004920.3 expression 12.06±3.33 was significantly lower in patients with gastric adenocar-cinoma with venous embolus than that without venous embolus(10.05±3.32,P=0.003),and was positively correlated with D-dimer(r=0.679,P<0.001).The early apoptosis rate of AGS cells in AC004920.3 overexpression group(33.45±3.90)%was higher than that in untransfected group[(12.13±1.54)%,P<0.001)]and transfection negative control group[(13.18 ±1.55)%,P<0.001].The proliferation of CCK showed that OD value of AC004920.3 overexpression group(2.89±0.17)was lower than that of untransfected group and transfection negative control group(3.20±0.21,P=0.003;3.28±0.18,P=0.001).The scratch area of AC004920.3 overexpression group(3.54±0.12)was significantly larger than that of untransfected group and transfection negative control group(2.43±0.34,P=0.001;2.60±0.39,P=0.004).Conclusion The serum AC004920.3 expression in patients with gastric adenocarcinoma with venous embolus was lower than that without venous embo-lus,and was positively correlated with D-dimer.AC004920.3 promoted the apoptosis of AGS cells and inhibited the prolifera-tion and migration of AGS cells.
